Climb up the root-covered wall to the right to break into their room. As you jump down on them, you can land an assassination on one or two of the guards before picking a fight with the rest. Kill 'em all, then open a door to the south. Pull the lever in the next cavern and you'll get a lengthy camera pan that shows a bunch of switches falling into place. There are four switches that need to be hung on in order to open a door to the sarcophagus.
The switches, and even some poles that you need to get there, are on timers, so you'll need to be quick. Once you kill him, the other will run away. Catch up to him and then make your way to the next target, a doorway to the main puzzle. This "puzzle" is simply a climbing course that focuses on changing jumping direction while running up a wall. Use a bookcase to the southeast to get into Lorenzo's bedroom, and from the top of it, jump towards another golden painting to the west. You're going to want to shimmy across the edges of the room until you get to another bookcase with yet another painting above it.
